Abstract Amongst the various irnmunodiagnostic assays, ELISA has been reported to be the most sensitive and objective assay. Cellular antigen ELISA was developed and standardized for detection of antitheilerial antibodies. Cells from continuously growing T. annulata schizont cell lines were washed, fixed with gluteraldehyde, and used as cellular schizont antigen. Soluble schizont and piroplasm antigens were prepared by lysing and sonicating culture-derived schizonts and infected erythrocytes, respectively. ELISA was conducted using three antigens, separately, on same known positive serum samples. On comparison of the three antigens the assay detected highest antibody titres using piroplasm antigen. For detection of antischizont antibodies, the use of cellular schizont antigen in place of soluble schizont antigen improved specificity and sensitivity of ELISA significantly.
