Online published on 20 May, 2023. Abstract Silybum marianum (L.) Gaertn is a medicinally important herb that is famous in production ofliver protecting compound silymarin, an isomeric-mixture of flavonolignans and taxifolin. In this study, we report on the effect of plant growth regulators (PGRs) in diverse concentrations and combinations coconut water (CW) and different types of explantes (root, hypocotyls, cotyledonary leaf) on callusinduction and silymarin production. Free radical scavenging activity of silymarin extracted from calluscultures of these explants was investigated (Three types of explants were excised from ten days old invitro grown plantlets viz. root, hypocotyl and cotyledonary leaf and inoculated on MS (Murashige and Skooge, 1962) medium (MSM) enriched. Root explants cultured on MS medium enriched with 5.0 μMNAA + 2.5 μM BAP + 10% CW showed the best results for inducing the maximum amount of friablecallus as well as the higher level of production of silymarin and free radical scavenging activity incomparison to other PGRs and explants tested. Therefor, for further research on silymarin production in cell suspension cultures, we suggest using a similar protocol for induce soft and friable calli in S. marianum.
