Effect of Growth Regulators on Micro Propagation of Potato (Solanum tuberosum L.)

Abstract

The present study was conducted to know the effect of growth regulators on micropropagation of potato. Three potato varieties, viz. Kufri Himsona, Kufri Swarna and Kufri Jawahar were evaluated for their responses to in vitro propagation using MS and B5 basal medium with varying concentrations of 2IP, GA3 and NAA. Kufri Jawahar exhibited the maximum stem elongation (10.48cm) in MS media consisting of 2IP, GA3, NAA 0.5, 0.5 and 0.1 respectively followed by B5 media containing 2IP, GA3, NAA 0.2, 0.5 and 0.5 respectively. Shoot regeneration from the nodal explants of three different potato cultivars was observed to be ranging from 74% to 96.4%. Maximum regeneration percentage (96.4%) was found to be on MS basal medium with 2IP (0.5 mg/l) + GA3 (0.1 mg/ml) in Kufri Himsona. Highest number of nodes were observed incase of Kufri Jawahar (8.1) in B5 basal media consisting of 2IP, GA3 and NAA 0.5, 0.5 and0.1 respectively followed by Kufri Himsona (7.9) in B5 media consisting 2IP, GA3 0.5 and 0.1 mg/l respectively. Successful rooting was achieved by placing the shoots on half strength MS basal medium. The present study clearly shows that different cultivars respond differently towards tissue culture conditions.
