Agroinoculation with Cloned DNA Particles of Mungbean Yellow Mosaic Virus and Their Whitefly Transmission in Black Gram (Vigna mungo L.)

Abstract

Mungbean yellow mosaic disease is one of the most serious biotic stress of pulses infecting major leguminous species including black gram (Vigna mungo (L.) Hepper). Present study was undertaken to compare the laboratory screening protocol with whitefly (Bemisia tabaci) transmission of MYMV. Two Agrobacterium viral constructs harbouring partial MYMV genome(s) of two different MYMV isolates namely VA221 (KA30 DNAA+ KA22 DNAB) and VA239 (KA30 DNAA+ KA27DNA B) were used for agroinoculation. Agroinoculated plants did not show any mosaic symptom even after one month of inoculation. Besides plants inoculated with VA239 viral construct showed stunted appearance. However both the constructs were detected by PCR amplification, where viral DNA was observed 7 and 15 days after inoculation with VA239 and VA221 viral constructs respectively. Whiteflies with three acquisition access period (AAP) viz., 24, 48 and 72 hr successfully transferred the virus from field infected plants but not from agroinoculated plants.
