A rapid and efficient protocol for isolation of genomic DNA from Chenopodium album and Lepidium latifolium has been described. The method involves modified CTAB extraction, employing addition of polyvinylpyrollidone (PVP) in CTAB buffer, high salt concentration, and successive long-term incubation in Chloroform: lsoamyl alcohol and R Nase treatment, with all of the steps carried out at room temperature. The DNA contained minimum contaminating metabolites. The genomic DNA samples,isolated by this methodfrom both the species showed distinct amplification with desired molecular weight. Reproducible amplification was observed in PCR reactions.
