Lactic acid bacteria are largest group of probiotic bacteria used in pharmacy. In present study, L. acidophilus, isolated from milk samples were identified and characterized using morphological, biochemical and molecular tools. All the bacteria were found to be Gram-positive and rod-shaped which were grown in presence of bile salt, and fermented sugars like maltose, lactose, sucrose and glucose at the temperature of 37 ºC. L. acidophilus survived in 4.5 % of NaCl concentration and at lower pH 5. PCR assay of the 22 L. acidophilus isolates led to the detection of 23SrRNA genes of molecular weight 575bp.
