Isolation, Screening and Identification of Rhizosphere Bacteria from Curcuma longa L.

Abstract

The productivity of Turmeric (Curcuma longa L.) is severely hampered by rhizome rot disease caused by Pythium aphanidermatum. Present study was undertaken to isolate, screen, and identify biocontrol agents from rhizosphere of this crop.. Nine bacterial isolates were obtained, which were evaluated antagonistic activity using the dual culture assay. Out of these, isolate Endo-11 for showed highest inhibition (85.88%) against P. aphanidermatum, followed by Endo-10 (82.88%) and Endo-14 (77.50%). Molecular identification using 16S rRNA gene sequencing identified the three most potent isolates as Bacillus siamensis JSS (Endo-11; GenBank: OL989163), Bacillus tequilensis SSJ (Endo-14; GenBank: OM085661), and Bacillus licheniformis SSJ (Endo-15; GenBank: OM184285). These findings suggested that B. siamensis JSS is promising bacterium for eco-friendly management of turmeric rhizome rot.
