Plant regeneration, genetic transformation and gene expression in In vitro tissues of cauliflower (Brassica oleracea L. var. Botrytis)
Abstract
Plant regeneration and Agrobacterium-mediated gene transfer studies were carried out in cauliflower (Brassica oleracea L. var. botrytis cv. Pusa Snowball K1). Higher frequency of shoot regeneration was obtained in cotyledon (68%) and hypocotyl (84%) explants on MS medium supplemented with 2.0 mg/l BAP + 0.5 mg/l IAA. Addition of 0.05 mg/l IBA in nutrient medium promoted regeneration of roots. Agrobacterium tumefaciens, strain LBA 4404 was used for genetic transformation studies. Transformed cells were found to be able to grow on selective medium. Successful genetic transformations were confirmed by β-glucuronidase enzyme assay.
