Analysis of Genetic diversity in pigeon pea (Cajanus cajan L.) germplasm using simple sequence repeat (SSR) markers

Abstract

The aimed of present study was to evaluate the genetic diversity of 10 Pigeon Pea cultivars by Simple Sequence Repeat (SSR) or microsatellite marker. The genomic DNA of 10 pigeon pea genotypes were amplified with 10 SSR primers that produced 27 amplified band, out of which 18 band were polymorphic (66.66%). The number of fragment amplified per primer ranged from 2 to 4. Primer CcM 2977 generated maximum number of amplified band, out of which 2 band were polymorphic. Cluster analysis of pigeon pea genotypes were based on UPGMA method. Cluster analysis of 10 pigeon pea genotypes were classified in to two main group; single variety ICPL990 was placed in first group and rest 9 variety were placed in second group. The pair wise similarity values ranged from 0.80% to 92% and showed that genotypes AH 069 and ICPA203 were the closest with highest similarity value (92%), while genotypes ICPL990 and JBP110B showed minimum similarity value (80%). On the basis of present study and previous studies we can concluded that the presence of high genetic diversity among pigeon pea cultivars is an urgent requirement to incorporate divers genotypes in breeding programmes, which could be used for the developing core collection of pigeon pea germplasm for breeding purpose.
