Quorum sensing (QS) represents a sophisticated cell density-dependent communication mechanism that enables bacterial populations in plant-associated environments to coordinate collective behaviors and optimize survival strategies. This review examines the molecular architecture of major quorum sensing systems, including N-acyl homoserine lactone (AHL), autoinducing peptide (AIP), and autoinducer-2 (AI-2) systems, and their functional roles in regulating bacterial activities within the phytomicrobiome. We explore how plant growth-promoting rhizobacteria (PGPR) and biocontrol agents utilize quorum sensing to modulate beneficial functions such as phytohormone production, siderophore synthesis, and antimicrobial compound secretion. Conversely, plant pathogens, including soft rot and vascular pathogens, employ quorum sensing to synchronize virulence factor production and coordinate infection strategies. The ecological significance of quorum sensing in rhizosphere dynamics, including its role in biofilm formation, horizontal gene transfer, and plant-microbe signaling dialogues, is critically analyzed. Furthermore, we discuss emerging biotechnological applications, particularly quorum quenching strategies for biocontrol and their integration into sustainable agricultural practices. Finally, we highlight methodological advances in biosensor development and imaging technologies, alongside the need for systems-level understanding of quorum sensing networks in complex plant-associated environments. Understanding and manipulating bacterial quorum sensing systems holds considerable promise for developing eco-friendly approaches to enhance crop productivity, manage plant diseases, and reduce dependence on chemical pesticides in modern agriculture.
