In Vitro Study On Plant Multiplication Of Sugarcane (Saccharum Officinarum L.) Variety Co 0118 And Co 0238 Under NaCl And Mannitol Stress

Abstract

A comparative in vitro study to analyze the salinity tolerance in two sugarcane variety viz. Co 0118 and Co 0238 were carried out on a selective Murashige and Skoog (MS) medium containing NaCl (0 mM, 50 mM, 100mM, 150mM, 200mM and 250 mM) and Mannitol (0, 80, 160, 200, 240, 320 mM) supplemented with 0.5 mg BAP and 0.5 mg Kinetin respectively. In this study in vitro performance of sugarcane (Saccharum officinarum L.) plants tolerant and sensitive to ionic and osmotic stress was achieved using shoot tip culture with five replicates each, in a completely randomized design (CRD). Experiment conducted to evaluate the plant multiplication rate in stress induced medium at three different durations of 7, 14 and 21 days, each repeated 5 times, was found significant at morphological level after 2 weeks only. In between these two varieties, Co 0238 was found more resistant than Co 0118 in both (NaCl and Mannitol induced) the medium. NaCl at 150 mM and Mannitol at 160 mM concentration was found as critical point in regulating the plant growth negatively, and concentration above these, plant shows a continues decline in its growth and development. In conclusion, NaCl and Mannitol both causes inhibition of plant multiplication in both the cultivars of sugarcane at particular concentration. The analysis of variance for all the parameters (number of shoots inoculated, number of shoot regenerated and shoot length) were found to be significant at P<0.05%.
