Abstract

Stripe rust caused by Puccinia striifromis f. sp. tritici is one of the major diseases of wheat causing substantial yield losses worldwide. P. striiformis pathotype (78S84) was collected from the Regional Station, ICAR-Indian Institute of Wheat & Barley Research, Flowerdale, for the molecular characterization. Molecular characterization of pathotype of Pst (78S84) was done using mitochondrial cytochrome oxidase (COX-1) markers. A partial region of COX-1 (334bp) of pathotype of P. striformis was amplified from genomic DNA, cloned into pTZE57R/T vector and capillary sequenced. Nucleotide sequence analysis (GenBank Accession No. MN-718665) revealed unique synonymous nucleotide changes of P. striiformis (78S84). Phlyogenetic analysis of P. striiformis (GenBank Accession No. HQ012475 and HQ147441), but (78S84) COX1 showed close relationship with other pathotypes of had differences from other Puccinia spp.
