Fingerprinting of rice (Oryza sativa) cultivarsusing Inter simple sequence repeat markers

Abstract

Variety identification has great significance from seed production, breeding as well as intellectual property rights point of view to ensure quality of seed. The present investigation was undertaken with an objective to identify distinguishable molecular markers to establish fingerprints of rice (Oryza sativa L.) varieties released from Indira Gandhi Krishi Vishwavidyalaya, Raipur. Eight Inter simple sequence repeat (ISSR) markers were employed for fingerprinting in four rice varieties namely Karmamahsuri, Danteshwari, Samleshwari and Mahamaya. A total number of 67 alleles were obtained using eight ISSR markers (UBC 842, UBC 810, UBC 812, UBC 834, UBC 808, UBC 873, UBC 841 and ISSR 2) were found to be the most informative primer sets with an average of 8.37 alleles per primer. The number of alleles amplified for each primer ranged from 13 to 6. Polymorphic information content (PIC) for these primers ranged from 30.00 to 38.88. The gene diversity for eight ISSR markers ranged from 0.815 (UBC 873 and ISSR 2) to 0.904 (UBC 804) with an average of 0.851. Banding pattern based on these primers produced unique DNA fingerprint for each variety. DNA fingerprints of rice cultivars by means of ISSR provided meaningful data, which can be extended by additional microsatellite markers. The data obtained can be used for the protection of plant genetic resources.
