Studies the sexual and In-vitro propagation of Ziziphus species

Abstract

Seeds of Chinese date (Ziziphus jujuba Mill.) without coat gave good result with 76–88% germination. Embryos from Chinese Spinosa (Ziziphus spina-christi) were growing successfully with good results. It was found that 80–91% of embryos were growing after the heat-cold and heat stratification. In both years, 2006 and 2007, heat-cold stratification has given the best results followed by heat stratification. In MS media, 40 segments (explants) of ‘Lang’ were used with different duration of disinfection of segments with HgCl2 (7, 9, 11 min.). It was observed that 65%, 50% and 47.5% of segments were infected in 7, 9 and 11 min. of HgCl2 treatment, respectively. Segments sprouted without any infection were 15%, 25% and 22.5% at 7, 9 and 11 min. of HgCl2 disinfection, respectively. About 20%, 35% and 30% of segments at 7, 9 and 11 min. of HgCl2 disinfection did not produce any sprout, but had not been infected. In DKW, 25, 36 and 36 segments of ‘Lang’ were used with 7, 9 and 11 minutes of the disinfection with HgCl2, respectively. About 32%, 33.3% and 25% of segments sprouted without any infection at 7, 9 and 11 minutes of HgCl2 disinfection, respectively. Segments found without any infection and sprouts were 16%, 25% and 25% at 7, 9 and 11 minutes of HgCl2 disinfection, respectively. Jujube cultivar ‘Lang’, DKW media with 9 min. of disinfection of HgCl2 is the most favorable.
