Morphological, Biochemical and Electrophoretic Evaluation of Hulled and Hull-less Varieties of Barley (Hordeum vulgare)

Abstract

Barley (Hordeum vulgare) is one of the cereal crop among top five major crops of the world which is widely used for food, feed stock, malting, brewing and distilling industry. In present study one hull-less (NDB 943) and two hulled (NDB 1173 and RD 2552) barley varieties were studied on the basis of their seed morphology, different physiological and biochemical parameters. Under morphological variation, days of 50% flowering and maturity were found higher in hulled varieties as compared to hull-less, while the number of grains per ear, test weight, biological yield per plant, grain yield and harvest index were higher in hull-less variety. Total soluble protein content in seeds of hull-less barley was found higher (9 mg/g) than in hulled varieties (6 mg/g in NDB 1173 and 7.12 mg/g in RD 2552). Protein profiling of seeds by SDS-PAGE showed one extra band (12 kDa) in hull-less variety i.e. NDB 943, which was not observed in hulled varieties i.e. NDB 1173 and RD 2552.
