Identification of Cotton (Gossypium herbaceum L.) Genotypes using Electrophoretic Techniques

Abstract

The present study was carried out to detect the polymorphism among fifteen genotypes of cotton (Gossypium herbaceum L) by SDS Polyacrylamide Gel Electrophoresis (PAGE) of total seed protein and isozymes (peroxidase and polyphenol oxidase) from fifteen days old cotton seedlings. Relative interspecific similarities of protein and isozyme patterns were estimated and cluster analysis was performed. Total 19 bands were observed with Rm values ranging from 0.137 to 0.935 through SDS-PAGE whereas peroxidase and polyphenol oxidase showed 6–7 isoforms with Rm values ranging from 0.242 to 0.866 and 0.399 to 0.903, respectively. Two main clusters were observed in the dendrogram of SDS-PAGE and Native PAGE. In conclusion, SDS-PAGE was found to be more effective for cultivar identification as compared to isozyme study.
