Analysis of genetic relationship of Pandanus tectorius accessions of Andaman and Nicobar Islands using rapd and issr markers

Abstract

Eighteen phenotypically superior accessions of Pandanus tectorius, identified from different locations of the Andaman and Nicobar Islands, were subjected to genetic diversity analysis using 20 polymerase chain reaction (PCR) markers (10 random amplified polymorphic DNAs (RAPDs) and 10 inter simple sequence repeats (ISSRs)). The percentage of polymorphic bands detected by ISSR and RAPD was 80.23 and 80.55%, respectively. RAPD primers OPF 04 and OPT 16 gave 100% polymorphism. Among ISSR primers, ISSR 11 and AMY24 gave 100% polymorphism. Clustering pattern of accessions of Pandanus remained more or less similar in RAPD and combined data of RAPD and ISSR. Present study confirmed very low level of genetic variation within the species even though it is exposed to a wide range of environmental conditions across Andaman and Nicobar Islands, India.
