Rumen Microbial Enzyme Activities on Transinoculation of Rumen Contents Between Cattle and Buffalo

Abstract

Transinoculation of rumen contents was studied on microbial enzymes of adult male flstulated cattle and buffalo, 4 each, fed wheat straw-groundnut cake diet (10–12% CP). After having been adapted the animals on test diet, rumen liquor samples collected at 0,3 and 6 hr post-prandially during3 stageslike pre-exchange, exchange and re-exchange of rumen contents of cattle and buffalo,.were differentially centrifuged to obtain protozoal and bacterial rich fractions. Extracts prepared out of such individual microbial fraction were assayed for glutamic acid dehydrogenase (GDH), glutamate oxalacetate transaminase (GOT) and gliita-mate pyruvate transaminase (GPT) besides cellulase in mixed culture of rumen liquor. The activities of intracellular proteosynthetic enzymes viz., bacterial GDH and protozoal GOT were induced (P < 0.05) by transinoculation. However, both the aminotransferases (GOT, GPT) In protozoa exhibited higher activities than those of bacterial enzymes while reverse was the trend for GDH activity. Re-exchange of rumen contents augmented (P< 0.05) the activity of ruminal cellulase compared to that of pre-exchange, though ruminant species did not vary for such enzymic activity. It may be inferred that through the process'of transinoculation of rumen contants explicitly and selectively (bacterial GDH and protozoal GOT) improved the activities of ruminal intracellular proteoanabolic enzymes, however the activity of extracellular cellulase was solely induced In mixed culture of the rumen.
