Determination of Enzyme Activities of Lignocellulolytic Strains in Solid Substrate Fermented Products

Abstract

CMCase, FPase, Xylanase and laccase were determined in solid substrate fermented (SSF) product and by cultivating Coprinus fimetarius (386), Phanerochaete chrysosporium (ME-446) and Streptomyces cyaneus (MT-813) in basal medium providing CMC, filter paper, xylan and wheat straw as carbon source for 16 d in order to assess the lignocelf ulose degrading efficiency of the strains. CMCase activity of C.fimetarius and P. chrysosporium increased in CMC substrate and in fermented product Filtrate. Similarly, on wheat straw as substrate, the activity increased regularly fori”, chrysosporium and S. cyaneus. Using C fimetarius on Wheat straw and S. cyaneus on CMC, the activities started declining after 10d of growth, but CMCase in SSF with S. cyaneus improved till 16th day. P. Chrysosporium and C. fimetarius exhibited greatest FPase activities on filter paper and during the SSF, respectively. The activity on wheat straw increased throughout for C. fimetarius and S. cyaneus while for P. chrysosporium, it first increased upto 6th day then declined at day 16. Usually xylanase activity was greater in wheat straw than on xylan in basal medium after 6-8d. The average ‘ activity had a tendency to increase in all trials in liquid or solid state. The maximum xylanase was released by P. chrysosporium using xylan as carbon source in basal medium while SSF product S. cyaneus showed highest activity. The laccase activity of C. fimetarius increased and then decreased after 14 d but for P. chrysosporium, it decreased while S. cyaneus showed downward trend after 12 d using wheat straw as substrate in liquid cultures. In case of filtrates from SSF product, the laccase activity increased till 16th day for P. chrysosporium and 5. cyaneus while C.fimetarius followed the same trend upto 14th day and decreased.
