Abstract

Different concentrations of hormones and their combinations were applied to most commonly used explants viz; hypocotyl and cotyledon cultured on MS medium. Swelling and callus formation was more frequent in cotyledon explant. The frequency and amount of callus was higher in treatment IAA (0.2 mg−1) +KIN (2.0 mg−1). This treatment was applied to different genotypes of B. Juncea. There was significant difference for days to callus induction, fresh and dry weight of callus. The earliest callus induction was observed in geotype T 59 i.e. 7819g and 0.654g respectively, at 45th DAI. All the genotypes regenerated shoots. In sub-culturing, genotypic difference were also significant for fresh and dry weights of callus. But trend of callus growth was different. Both fresh and dry weight of callus was observed maximum in genotype RH 781 Le. 5.498g and 0.564g, respectively. Shoot regeneration was not observed in any genotype in sub-culturing though all chemical and physical conditions were same.
