Abstract

NADP+- dependent isocitrate dehydrogenase was extracted and purified from 48 old germinated blackgram, Vigna mungo (L.) Hepper by a procedure including successive purification steps of homogenization, heat treatment, ammonium sulphate fractionation, ion exchange chromatography on DEAE-cellulose and affinity chromatography on Blue Sepharose CL-6B. In ion-exchange chromatography, the starting enzyme activity was resolved into four different enzyme peaks (isozymic forms) corresponding to elution buffer containing 25, 50, 75 and 100 mM KCl, respectively.
