Abstract

In animal agriculture, additional genetic selective advantages are gained when genetic enhancements can be made in already known superior genotypes. To gain this advantage, adult, genetically enhanced donor cells must be obtained. There are limited studies using adult transgenic donor cells in the nuclear transfer procedure. Recently, it has been shown that adult somatic cells could remain totipotent for cloning after long-term culture suggested the feasibility of genetic manipulation. Skin piece was obtained from ear pinna of elite Sahiwal cattle and the present study was undertaken to obtain cell lines from the ear skin biopsies. Various parameters like cell proliferation rate, chromosome number, apoptosis, TNF-α, IL-6, activity of caspases, LDH etc. were evaluated at every third passage till fifteenth passage. The results proved that culture was devoid of cytotoxicity and could be maintained till fifteenth passage, which can be further used for genetic manipulation/nuclear transfer studies.
