Effect of Different Carbohydrates Sources on the Formation of Cormlets of Economic Important Plant: Gladiolus Pacifica

Abstract

An efficient in vitro protocol for mass propagation of Gladiolus was developed. Basal portion of the leaves responded readily in culture. The explants formed regenerable callus when cultured on either NAA (2, 1, 0.4 mg/l), BAP (0.5, 1 or 2 mg/l). For shoot induction one gram (fresh weight) of callus, maintained in callus initiating medium with different concentration taken as sample (sucrose, fructose, glucose, mannitol), The highest number of shoots was found to be regenerated in medium with 3% from one-month-old callus maintained in medium with 0.4 mg/l NAA. Shoots cultured on half strength MS medium with 3% sucrose produced roots in 100% culture within two weeks. Rooted shoots produced cormels within eight weeks in the same medium and maximum number (12) of cormels was found to be produced in MS medium with 2 mg/l 3% sucrose in liquid medium and also in 6% fructose and 6% glucose. We observed highest number of cormel formation in medium with 6% fructose and glucose. In this research we have refine the large scale propagation protocol for Gladiolus.
