Drug Release Method by HPLC for Tamsulosin Hydrochloride 0.2% and Tolterodine tartrate 0.2% Combination Pellets

Abstract

In this study, a high performance liquid chromatography method was developed for the determination of Tamsulosin hydrochloride 0.2% and Tolterodine tartrate 0.2% Combination With U.V detector, The chromatographic system consisted of a Inertsil ODS 3V (4.6 mm X 150 mm) that contain 5μm packing column. Gradient method was used. 30.5ml perchloric acid with 95 ml of water and 10.5gm sodium hydroxide, made up to 1000ml with water and homogenized as mobile phase. The flow rate is about 2.0ml per minute and effluent was monitored at 220nm. Tamsulosin hydrochloride eluted at about 3.9min and Tolterodine tartrate eluted at 13.6min, no integrated peak from excipient used for preparation of dosage form. The method was linear over the range of 100–500μg/ml. The appropriate dissolution conditions were also determined and applied to evaluate the dissolution profile of Tamsulosin hydrochloride and Tolterodine tartrate pellets. Based on the stability and basic nature of the drug, dissolution experiments were conducted in 900ml of pH 6.8 phosphate buffer with Basket, rpm; 100. Dissolution profile proved to satisfy the specification. So proposed method was Applied Successfully for the determination of Tamsulosin hydrochloride 0.2% and Tolterodine tartrate 0.2% Combination Pellets.
