Abstract

Toll like receptor (TLR) expression was evaluated in selected tissues of pig (lymph node, spleen, kidney, heart, stomach, lung, intestine, liver and muscle) using one-step reverse transcriptase polymerase chain reaction (RT-PCR). The present study confirmed the expression of TLRs obtained with the successful amplification using TLR specific primers for porcine. Expression of TLR-1, 2, 4 and 10 were observed in many tissues tested and among the tissues analyzed muscle and small intestine had least or no expression. Expression of TLR-5 was observed in all except muscle tissue, TLR-6 expression was observed in spleen, brain, heart, lung, lymph node and liver and TLR-7, 8 and 9 expression were observed in spleen, brain, heart, lung, lymph node, liver, stomach and kidney. The amplicons were subjected to restriction enzyme digestion using HinfI, DedI, HaeIII and AvaII for different amplified PCR products. This simple and rapid method can be used to study the expression for TLRs.
