Environmental mastitis due to Klebsiella pneumoniae is one of the major concerns in buffalo dairy herds with respect to antibiotic resistance. To alleviate the antibiotic resistance, an alternate therapeutic approach was undertaken. For this purpose, five lytic bacteriophages (KP1 to 5) with a host range of 82–90% individually and 100% collectively were characterised biophysically and were stable at different temperatures and pH ranges. The morphology of KP1 revealed that the phage belong to the family Siphoviridae and order Caudovirales and molecularly the nucleic acid was found to be DNA and got digested by the restriction endonucleases. The therapeutic evaluation was studied using Swiss albino mice (control, infected and treatment). Under histopathology, the mammary tissue sections from infected group revealed inflammatory changes with extensive neutrophilic infiltration, whereas the sections from treatment group revealed mild inflammatory reaction which indicated the therapeutic efficacy of lytic bacteriophages in the treatment of mastitis.
