Hematological Changes in CPDA-1 Anticoagulant Containing Blood Bags: A Study of Blood Storage

Abstract

The study was aimed to compare hematological changes in stored blood in CPDA-1 containing blood

bags, in relation to different time periods ranging from 1 to 35 days. All blood donors were male,

voluntary or replacement and were tested negative for HIV 1&2, HCV, HbsAg, Syphilis and Malaria.

The hematological parameters were measured using the Horiba ABX SAS, fully Auto Hematology Analyzer (France), while Wintrobe`s method was used for estimation of ESR. There was moderate decrease in hemoglobin concentration and PCV, while platelets count and total WBC count were markedly reduced. Comparison of mean value of day 1 versus day 35 revealed that the granulocytes were drastically reduced. ESR values increased from 6.2 to 10 mm/hr. Differential count of lymphocytes % increased from 32.8 to 69.3, monocytes % increased from 8.1 to 19.2 but granulocytes % decreased from 59.1 to 11.2. There is mild increase in MPV and RDW. The transfusion of blood stored for different periods of time was found to have reducing effect on hemoglobin and PCV. Therefore old blood should not be used for the treatment of anemia. Rapid degeneration of leukocytes could lead to immunodulation related to blood transfusion. Whole blood should be leukodepleted before storage and it must be used within one week. There will be no increase in WBC and platelet count if blood is transfused after 24 to 36 hours of storage at 4 to 6°C.
