The present work was undertaken to amplify the hexon gene specific EDS -76 virus isolated in Namakkal and its RFLP pattern. The isolate was inoculated into 11-day old embryonated duck eggs. The harvested allantoic fluid was subjected to Haemagglutination test (HA) and the presence of virus confirmed by neutralization of HA activity by using standard hyper immune serum. Hexon gene of EDS-'76 virus was amplified by polymerase chain reaction. The amplicon was subjected to restriction enzyme analysis. BstYI, Taq I and Hind III restriction enzymes were used and the fragment patterns were studied in 1.5% agarose gel electrophoresis. After the digestion with BstYI, TaqI, Msp I, three bands of size approximately 0.86, 0.63, 0.41 kb were obtained by BstYI digestion, two bands of size approximately 1.45, 0.45 kb were obtained by TaqI digestion, three bands of size approximately 1.56, 0.22, 0.12 kb were obtained by Msp I digestion.
