Intact mesophyll protoplasts from gladiolus leaves were isolated by digesting leaf material for 10 hours in Onozuka R-lO cellulase 1 per cent + pectinase 0.5 per cent with CPW salt solution without copper sulphate. Peeling of epidennis was better than shredding and scoring treatments given to the leaf material before preplasmolysis. leaf material in enzyme solution which were kept on rotary shaker gave more number of intact protoplasts than the stationary enzyme digestion.
