In vitro plant regeneration of Dianthus caryophyllus cv. Alas Red through callus culture

Abstract

In vitro shoot bud regeneration was achieved from callus derived from stem and leaf explants of Dianthus caryophyllus cv. Alas Red on Murashige and Skoog basal medium supplemented with 1-2 mg/l BA and 0.25 mg/l 1-naphthylacetic acid (NAA). Frequency of regeneration was better in calli derived from the leaf as compared to the stem explants. The average number of shoot buds/culture were 40.6 ± 0.3 and 26.2 ± 0.4 in leaf and stem explants respectively. The in vitro grown shoots were rooted on 1/2 strength basal MS salts supplemented with IBA (0.2 mg/l). Higher concentrations of IBA (0.25-0.5mg/l) inhibited the rooting response. The micropropagated plantlets grew and flowered normally.
