In vitro polyphenol oxidase activity and polyphenol pattern in shoot tip cultures of grape

Abstract

Catecholase and cresolase enzyme activity pattern varies with the duration of cultures. During first 15 days of culture establishment, an increasing pattern in enzyme activity was observed, while later on, activity starts declining at each subsequent interval. Consequently, polyphenol content also increased and decreased with enzymes activity pattern. Though the cresolase activity pattern was similar to catecholase but its level was much lower than catecholase. PPO activity and polyphenol content varied significantly among different genotypes. A very high positive correlation (r=+0.92) between PPO activity and polyphenol content establishment the fact that it is also responsible for in vitro synthesis and accumulation of phenolic compounds.
