Rapid in-vitro propagation of Rosa hybrida through axillary buds

The most suitable culture medium was MS medium on which rapid establishment and differentiation was observed. Among the different concentrations of the 3 phytohormones used individually and in various cambinations, early establishment and differentation of axillary buds was observed in MS medium supplemented with 2 mg l⁻¹ and 4 mg l⁻¹ kinetin and 2 mgl⁻¹ and 4 mgl⁻¹ BAP individually and also in combination with 0.1 mgl⁻¹ NAA. Induction of multiple shoots was maximum in MS medium supplemented with 4 mgl⁻¹ BAP and decreased with the concentration of the BAP supplementation. However, only single shoots developed on cultures supplemented with kinetin. Though longest shoots with the highest number of leaves developed on medium supplemented with 2 mgl⁻¹ kinetin followed by 2 mgl⁻¹ BAP, they failed to develop roots on the rooting medium. The shoots regenerated from 0.25 mg l⁻¹ BAP and 0.25 mg l⁻¹ kinetin supplemented media responded most efficiently to rooting and subsequent survival after the transfer to the greenhouse.
