Studies on in-vitro plantlet production in gladiolus cv. Friendship

Abstract

In-vitro studies were conducted to find out the plantlet production in gladiolus cv. Friendship. The shoot tip explants were cultured on MS medium supplemented with 2,4-D, NAA, kinetin and BAP in various combinations. Exogenous supplementation of cytokinin and auxin was found to be essential for shoot and root regeneration respectively. The shoot regeneration increased with increase in kinetin and BAP. Similarly, root regeneration increased with increase in NAA and 2,4-D. The highest shoot regeneration was observed only in the medium supplemented with cytokinin. The highest root regeneration was observed in medium supplemented with auxin. Among the combinations tried, kinetin (4 mg/litre) gave highest shoot regeneration and NAA (4 mg/litre) gave highest root regeneration. The best combination also induced higher shoot number, shoot weight, root number and root length.
