Standardization of culture conditions for in-vitro germination in subtropical peach (Prunus persica) cv. Flordasun

Abstract

The best basal media for embryo germination and root and shoot growth was found to be B5 (macro) + MS (micro + organics). Sucrose level (30 g/litre) was found most effective for embryo germination as it gave earliness in root and shoot emergence. The embryo germination was markedly triggered by plant bioregulators and treatment comprising BAP (1.5 mg/litre) supplemented with GA3 (1.0 mg/litre) resulted in highest germination. The period required for emergence of root and shoot was also reduced by plant bioregulators treatment and minimum duration for root and shoot emergence was recorded in the treatment comprising BAP (1.5 mg/litre) supplemented with GA3 (1.0 mg/litre). The germination of embryo was significantly better with incubation in 12/12 hr light and dark cycle.
