Studies on in vitro shoot regeneration in grape cvs. Perlette and Flame Seedless

Abstract

Perlette and Flame Seedless are the important grape cultivars of Punjab. In vitro shoot regeneration studies were carried out in these cultivars. Nodal segment (2–3 cm) and young leaves at three different stages (stage I, II and III) were used as explant. Maximum explant (nodal segment) establishment in both the cultivars was obtained in April. The highest shoot regeneration frequency in Perlette and Flame Seedless was obtained on MS media fortified with BAP (1 mg l−1) + kin (1 mg l−1) and BAP (1 mg l−1) + kin (0.5 mg l−1), respectively. For nodal segments, increase in either of the cytokinin (BAP or kin) beyond 1 mg l−1 in the medium resulted into subsequent reduction in shoot regeneration percentage. Shoot regeneration from leaves was achieved only in cv. Flame Seedless and stage I leaf were the most responsive. Maximum shoot proliferation in Perlette and Flame Seedless was obtained on MS medium supplemented with BAP (1 mg l−1) + kin (2 mg l−1) and BAP (1 mg l−1) + kin (1 mg 1−1), respectively.
