The effects of amino acids (glutamine, serine, proline, glycine and valine) and additives (PVP, sodium citrate, activated charcoal) on plant regeneration from cotyledon and embryonal axes explants of peanut were studied. Cotyledon showed better response than embryonal axis explants. The combination of BA (1.5mM), NAA (1.0mM), PVP (25mM) with L-glutamine (15mM) enhanced the maximum number of shoots (36.2 shoots/explants). The multiple shoots were separated and transferred to shoot elongation medium containing BA (1.0 mM) and GA3 (0.5mM). The elongated shoots were rooted on MS medium containing NAA (1.0mM). A high percentage of (97.5%) plants were successfully hardened.
