Abstract

Plasmid DNA of Pigeonpea proteinase inhibitor was isolated by alkaline lysis method. The insert of PPI was used as heterologous probe for Lentil proteinase inhibitor gene Isolation. The etiolated seedlings were used for genomic DNA isolation. In genomic analysis of DNA one band of high molecular weight DNA was observed along with RNA band at the bottom of gel. The purified genomic DNA digested with EcoRI and bound to nitro cellulose membrane. The radioactive probe used for hybridization with nitro cellulose membrane. The probe DNA was efficiently bound to membrane. The autoradiogram showed the presence of protease inhibitor gene in Lentil. The radio labeled heterogeneous pigeon pea proteinase inhibitor used as probe to isolated proteinase inhibitor gene from Lentil.
