Isozyme analysis is a powerful biochemical technique with numerous applications in plant physiology. It has long been used by geneticists to study the population genetics of fish, mammals, insects, nematodes and higher plants. Malate Dehydrogenase and Peroxidase isozyme were used to identify the variation among popular wheat cultivars of India. Malate Dehydrogenase and Peroxidase isozyme are Ubiquitous enzymes, easily extracted and stable in cell-free preparations. The particulate and soluble fractions of cell-free extracts from leaves of eight different varieties of wheat plants were examined electrophoretically for isozymes of malate dehydrogenase and Peroxidase. Distinct isozyme patterns were observed for each variety. There were some isozymes in different plant extracts with equal electrophoretic mobilities. But there was some isozyme band that was common to all to all plants. In this Paper an attempt has been made to study diversity in Malate Dehydrogenase and Peroxidase for understanding the species interrelationship and variation among wheat varieties for their biotic and abiotic stress tolerance, to provide further indications about their genetic relations.
