The response to different growth substances varied depending on the explant type in Nicotiana plumbaginifolia. The leaf explants form ad comparatively more shoot buds in response to kinetin whereas the stem segments differentiated more shoot buds on MS medium+kinetin+IAA. The stelar tissue of the stem segments was observed to be inhibitory for shoot bud differentiation at lower kinetin levels (0.1–0.5 mg/1). GA, at lower levels (0.5 mg/1) enhanced shoot bud differentiation in leaf explants but not in the stem segments.
