Chloroplasts of wheat, maize and spinach were isolated in two isotonic media i.e., sodium chloride and mannitol. Mannitol always proved to be superior i.e., than sodium chloride. In spinach nearly 72 to 80 per cent activity was associated with chloroplasts where as in wheat and maize chloroplasts associated carbonic anhydrase level was 38.0 and 10.7 per cent respectively, thus disproving the generalization that in C3 plants enzyme is present in chloroplasts and C4 it is associated with cytosol fraction.
