Abstract

Attempts were made to find out the physiological requirements for callus formation from the anthers of pigeon pea. The investigations suggest that the modified Murashige and Skoog liquid medium supplemented with 2, 4-D (2.0 mg/1) and kinetin (0.2mg/l) can be used to induce callus from the anthers of pigeonpea. The callus thus formed can be maintained on the same medium solidified with Agar (10.0 g/1). The ploidy analysis of the callus reveals the diploid nature, thus indicating the origin of the callus from the vegetative cells of the anther but not from the pollen grains. Genotype and physiological status of the material seems to play an important role in callus production of anthers.
