Abstract

Sucrose phosphate synthetase (SPS) a key enzyme which regulates the sucrose synthesis in the leaf, is labile in nature. In vitro extraction and delay in the assay leads to loss of enzyme activity. A few experiments were conducted to test whether casein could be used in the extraction buffer for stabilizing SPS activity. The results indicate that presence of 3% casein in the extraction medium had not only stabilized the SPS activity even at 15°C preincubation conditions but also resulted in over 100% increase in the activity over no casein control.
