Studies were undertaken to raise axenic cultures and axillary bud induction from field grown plants of 20–25 years old Acacia nilotica. Heavy contamination and release of leachates were the major problems in establishment of explants in vitro. Bud break in nodal segments was observed in MS and WP media supplemented with BAP (0.25 - 0.5 mg/1) and/or adenine sulphate (20mg/l). Use of antioxidant viz. ascorbic acid, citric acid and sodium suphate (100mg/l) checked the media browning due to leachate exudation during in vitro establishment. Frequency of outbreaks was more in explants implanted in true polarity than in reverse polarity.
