In order to investigate the presence of Cry1Ac gene, transgenic plants were analysed by Polymerase chain reaction (PCR). Leaf samples from five Bt cotton hybrids along with leaves from non Bt hybrids as control were used. PCR analysis showed the presence of Cry1Ac gene in all five Bt cotton hybrids tested. The quantitative Enzyme linked immunosorbent assay was conducted to estimate Cry1Ac levels in five Bt cotton hybrids for two consecutive years during 2005–2006 and 2006–07. The Cry1Ac protein content was high in leaves which ranged from 2.35 to 5.99 μg/g fresh weight followed by squares (0.86 to 2.00 μg/g fresh) and bolls (0.73 to 1.92 μg/g fresh weight), respectively at 55–85 days after sowing (DAS). The Cry1Ac protein content in all the plant parts decreased by 100 DAS and further reached to very low level by 130 DAS. The order of Cry1Ac insecticidal protein content in different plant parts were leaves > squares > bolls.
