DNA Fingerprinting using synthetic oligonucleotide probe in native and exotic chicken breeds

Abstract

The DFP patterns were developed by hybridization of Hinf I and Hae III digests of the individual DNAs from two chicken breeds viz. White Leghorn (WL) and Kadaknath (KN) with gama32_P labeled-oligonucleotide (GTG), as probe. The average number of DFP bands were 7.5±0.8 and 6.6±0.7 in WL and KN breeds, respectively. The corresponding values of proportion of shared bands were 0.64 and 0.42. Similarly, the values of probability of two individual birds to exhibit identical fingerprint were 7.3 x 10−4 and 2.8 x 10−5 in WL and KN, respectively. Higher band sharing rate and probability of identical fingerprints in WL as compared to KN indicated reduced variability of these microsatellite loci among the individuals of this breed. The present results showed the effectiveness of DFP analysis using a synthetic oligonucleotide probe in detecting the polymorphism among the chicken populations.
