The genome analysis studies, requires large sample size and at times, it becomes necessary to store the sample fot longer duration before it is being processed for DNA isolation. The genomic DNA extracted from fresh and stored blood of four poultry species namely chicken (Gallus domesticus), quail (Coturnix japonica), guinea fowl (Numida meleagris) and turkey (Meleagris gallopovo), was evaluated for quality and quantity. High quality intact DNA (260/280: 1.7–1.9) was extracted from both types of blood samples in all the species. Average yield of 300–400 μg genomic DNA per 50 μl of blood was obtained. Restriction endonuclease digestion and RAPD amplification of the extracted DNAs from either type of blood were reproducible, indicating that the qualityas well as quantity of isolated DNA is not affected by the long storage of blood samples and similar protocol can be used for DNA extraction from either type of blood.
