Randomly amplified polymorphic DNA polymorphism among native and exotic chicken breeds

Abstract

The randomly amplified polymorphic DNA (RAPD) markers were used fo detect polymorphism among four breeds of chicken i.e. White Leghorn (WL), selected for part period-egg production, White Plymouth Rock (WR), selected for early body weight Aseel (AS) and Kadaknath (KN)ythe native breeds. Three of the sixteen random primers screened yielded distinct polymorphicRAPD profiles. Of the total 23 fragments amplified from these three primers, about 65% of them showed polymorphism. The average number of amplified bands per primer ranged from 5.00 (WL) to 6.83 (AS). The proportion of polymorphic loci ranged from 20.0% (WR) to 33.3% (KN). The within breed genetic similarity, estimated from band frequency (WS) and band snaring (WS), did not seem to differ much neither for magnitude (WS = 0.85 to 0.93 and WF = 0.84 to 0.91) nor for trend with. each other. The exotic breeds showed higher genetic similarity within population in comparison to the native breeds. The between breed genetic similarity estimates from band frequency (BF = 0.55 to 0.74) were lower than the estimates fonn band sharing (BS = 0.77 to 0.96) in magnitude, but presented more or less similar trend. The native breeds showed maximum genetic similarity among themselves, while WL breed showed a least genetic similarity with other breeds. The dendogram constructed to show phylogenetic relationship among these breeds, also showed Similar trend.
