RAPD profiling of Bantam, White Leghorn and Bantamised White Leghorn birds

Abstract

Thirteen RAPD primers were tested on Bantam (BNT), White Leghorn (WLH) and Bantamised White Leghorn birds (BWLH). Out of 13 primers investigated, three (BG-1, PLANTAGO-01 and PLANTAGO-06) were polymorphic on sex-wise pooled DNA samples. The BG-1 amplified a sex (female) specific band (1285 bp) in all three groups. It also revealed 625 and 550 bp fragments specific to BWLH and WLH. The band sharing (BS) value between BWLH and BNT group was higher (0.4454) as compared to that between BWLH and WLH group (0.3880). The PLANTAGO-01 produced fragment (1014 bp) specific to BWLH and WLH and PLANTAGO-06 amplified fragments (790 and 832 bp) specific to BNT group. These two primers showed higher BS values between BWLH and WLH (0.6235 and 0.5235) as compared to BWLH and BNT groups (0.5346 and 0.4637). Phylogenetic analysis also revealed similar results.
