PCR-RFLP of chicken growth hormone gene in egg-type chickens
Abstract
In this study growth hormone polymorphism has been investigated in egg type chicken by PCR-RFLP. Three different profiles (BB, CC and AC genotypes) created by a combination of polymorphic MspI cut sites due to T+385C and A+119G SNPs in intron-1 were identified. The genotype amplicons (BB, CC) were sequenced and submitted in the genebank with accession nos. EF472953 and EF452679. The frequency of the heterozygous AC and homozygous CC genotypes was higher in pure layer chicken (RIR, WLh), respectively. In RIR × WLh cross layer chicken heterozygous AC genotype was found more frequently than the homozygous CC genotype. The homozygous AA genotype was not found in the pure layer chicken. The analysis of the two identified SNPs with growth traits showed that A+119G was significantly correlated with body weight at all ages in layer chicken. On phylogenetic analysis the Gallus gallus nucleotide sequences clustered into a monophyletic group closer to Meleagris gallopavo and farthest away from Cairina moschata.
