Genetic polymorphism and estimation of parental genomic proportion in a chicken population developed for marker assisted introgression of naked neck gene

Abstract

A resource population comprising of naked neck sire and F1 progenies (naked neck and normal feathered) were genotyped using 20 arbitrary short oligonucleotide primers and 10 microsatellite markers from chromosome 3. Six of the 20 random primers detected polymorphism between naked neck (Nana) and normal feathered (nana) F1 progenies. A total of 69 bands were amplified from six polymorphic RAPD markers and 19 of these (27.5%) were found to be polymorphic. Six of the 10 microsatellite markers showed polymorphism between naked neck (Nana) and normal feathered (nana) F1 progenies. Out of 23 alleles amplified from the 6 polymorphic microsatellite markers, 14 were polymorphic. Parental genomic proportion in F1 progenies was estimated as the genetic similarity between the sire and F1 progenies using the genotyping data with six informative microsatellite markers. The average genetic similarity and genetic distance between the sire and F1 progenies was 0.803±0.024 and 0.22±0.015, respectively. Present results revealed the effectiveness of microsatellite markers in detecting the polymorphism between the F1 progenies and estimation of parental genomic proportion of naked neck sire in F1 progenies.
